Widely-used fluorescent indicator for estimating intracellular pH. This membrane-permeant compound is modified by intracellular esterases to produce BCECF, a polar fluorescein derivative that is retained by cells. Intracellular BCECF is stable and has an efflux half-life of greater than two hours. Intracellular pH measurements are made by ratio imaging, which involves determining the pH-dependent ratio […]

Phenolic dyes with high pKas – such as 7-hydroxycoumarin (often called umbelliferone) and the more common 7-hydroxy-4-methylcoumarin – are not fully deprotonated and therefore not fully fluorescent unless the pH of the reaction mixture is raised to above pH ~10. Thus, substrates derived from these fluorophores are seldom used for continuous measurement of enzymatic activity […]

5-(and-6)-Carboxy-X-rhodamine, succinimidyl ester is an amine-reactive long wavelength rhodamine dye. Derivatives made with the amine-reactive carboxy-X-rhodamine (ROX), succinimidyl ester are widely used for oligonucleotide labeling and automated DNA sequencing applications. Conjugates of this dye have longer-wavelength spectra than the spectra of Lissamine rhodamine B conjugates, but somewhat shorter-wavelength spectra than those of Texas Red® conjugates.

Fluorescent probe and cell staining dye. Reacts with primary and secondary amine groups on proteins and other biomolecules. Cell permeable and covalently couples, via its succinimidyl group, to intracellular molecules. Due to this covalent coupling reaction fluorescent CFSE can be retained within cells for extremely long periods. Used for immunological studies to monitor the proliferation […]
