DM4
DM4 is a potent maytansinoid and potent inhibitor of tubulin. It binds to α/β-tubulin heterodimers and inhibits tubulin polymerization and microtubule assembly, leading to enhanced microtubule destabilization. This strong antimitotic activity results in suppressed microtubule dynamics, causing mitotic arrest and apoptotic cell death, making it valuable for studying acute myeloid leukemia (AML) therapies.
